Kwas sialowy i związane z nim enzymy metaboliczne okazały się ważnymi składnikami patofizjologii cukrzycy typu 2 (T2D). U ludzi i zwierząt z T2D występuje podwyższenie stężenia kwasu sialowego w surowicy. W niniejszym badaniu zbadano modulację poziomu ekspresji mRNA genów UDP-N-acetyloglukozamino-2-epimerazy / kinazy N-acetylomannozaminy (GNE) i neuraminidazy 1 (NEU1) w niektórych narządach szczurów z cukrzycą typu 2.
T2D indukowano za pomocą modelu fruktozy-streptozotocyny i osiem tygodni po wywołaniu cukrzycy oznaczano kwas sialowy we krwi i narządach (tkance tłuszczowej, mózgu, okrężnicy, nerkach, wątrobie, trzustce, mięśniach szkieletowych i śledzionie), a następnie oznaczono ilościowo mRNA poziom ekspresji genów GNE i NEU1 przez qPCR. Wyniki wykazały znaczący (P <0,05) wzrost poziomu kwasu sialowego w surowicy i we wszystkich wyżej wymienionych badanych narządach z wyjątkiem tkanki tłuszczowej i mięśni szkieletowych szczurów z cukrzycą w porównaniu z normalną grupą kontrolną.
Ekspresja genu GNE była zwiększona tylko w trzustce (1,8-krotnie) u szczurów z cukrzycą, podczas gdy nastąpiło zmniejszenie ekspresji genu w okrężnicy. Natomiast ekspresja genu NEU1 wzrosła w śledzionie (3,5-krotnie), mózgu (2,2-krotnie), wątrobie (1,9-krotnie), okrężnicy (1,5-krotnie) i nerkach szczurów z cukrzycą. Stwierdzono, że podwyższony poziom kwasu sialowego w narządach szczurów z cukrzycą, z wyjątkiem trzustki, może nie wynikać ze zwiększonej endogennej syntezy kwasu sialowego.
Jak aktywność sportowa wiąże się ze śmiertelnością, cukrzycą i stanem przedcukrzycowym dla różnych poziomów wskaźnika masy ciała?
W niniejszym badaniu zbadano związek aktywności sportowej z wynikami zdrowotnymi i czy związek ten różni się między subpopulacjami na poziomie wskaźnika masy ciała (BMI). Wyniki badań dotyczące aktywności sportowej porównano z innymi rodzajami aktywności fizycznej w czasie wolnym (PA). Zastosowaliśmy modele regresji proporcjonalnych hazardów Coxa do oceny związków między uprawianiem sportu i czterema innymi typami PA (jazda na rowerze, ogrodnictwo, wykonywanie dorywczych prac i chodzenie), z ryzykiem wystąpienia stanu przedcukrzycowego, cukrzycy typu 2 (T2DM) i śmiertelności z jakiejkolwiek przyczyny u 97 212 osób (58,4% kobiet; średni wiek: 46,5 lat) w kohorcie holenderskich Lifelines.
Wyniki zostały stratyfikowane według trzech poziomów BMI: zdrowej masy ciała (BMI 18,5-24,9 kg / m2), nadwagi (BMI 25,0-29,9 kg / m2) i otyłości (BMI 30,0 kg / m2 lub więcej). Aktywność sportowa wiązała się z mniejszym ryzykiem dla zdrowia, ale tylko istotnie w przypadku stanu przedcukrzycowego (HR = 0,86, 95% CI 0,81 do 0,92). W przypadku osób o zdrowej masie ciała aktywność sportowa wiązała się z największym zmniejszeniem ryzyka, przy znacznie niższym ryzyku wystąpienia stanu przedcukrzycowego (HR = 0,78, 95% CI 0,68 do 0,90) i śmiertelności z dowolnej przyczyny (HR = 0,79, 95% CI 0,65 do 0,96) .

Inne typy PA nie wiązały się z istotnie niższymi zagrożeniami dla zdrowia, z wyjątkiem jazdy na rowerze, w przypadku której stwierdzono znacznie mniejsze zagrożenie dla zdrowia osób z nadwagą. Nasze odkrycia pokazują, że aktywność sportowa wiąże się z mniejszym zagrożeniem dla zdrowia, zwłaszcza stanem przedcukrzycowym, ale wpływ różni się w zależności od poziomu BMI, przy czym najsilniejszy związek występuje u osób o zdrowej wadze. Udział w sporcie, wraz z jazdą na rowerze, prawdopodobnie będzie skuteczniejszy w zmniejszaniu zagrożeń dla zdrowia niż inne rodzaje PA.
miR-139-5p promuje neowaskularyzację w retinopatii cukrzycowej poprzez regulację fosfatazy i homologu tensyny
Patologiczna neowaskularyzacja siatkówki jest motorem postępu retinopatii cukrzycowej (DR). Niniejsze badanie miało na celu zidentyfikowanie mikroRNA (miRNA), które ulegają zróżnicowanej ekspresji podczas progresji DR, a także zbadanie specyficznego mechanizmu regulacyjnego tych miRNA w neowaskularyzacji siatkówkowej. Korzystając z zestawu danych z mikromacierzy i mysiego modelu z cukrzycą, określono, że miR-139-5p był znacznie zwiększony podczas progresji DR. Badanie in vitro ujawniło podwyższenie poziomu miR-139-5p zarówno w komórkach śródbłonka mikronaczyniowego siatkówki myszy (mRMEC) leczonych wysokim poziomem glukozy (HG), jak iw ludzkich RMEC traktowanych HG (hRMEC).
Nadekspresja miR-139-5p zwiększała migrację komórek, ułatwiała tworzenie rurek i zwiększała poziom białka czynnika wzrostu śródbłonka naczyniowego (VEGF) w hRMEC. Podczas gdy efekt angiogenny nadekspresji miR-139-5p został zatrzymany przez przeciwciało anty-VEGF.
